IL-18Rβ(白细胞介素18受体β,又称IL18RAP)是IL-18R复合物的信号转导链,也是IL-1R家族的成员。IL-18R beta 在 T 细胞、NK 细胞和树突状细胞上表达,但在间充质细胞中不常见表达。
人 IL-18R beta 蛋白的氨基酸序列与小鼠IL-18R beta蛋白同源性较低。
IL-18R beta是IL-18的受体。IL-18 通过与 IL-18 α 链 (IL-18Rα) 结合形成信号传导复合物。称为 IL-18 受体 β 链 (IL-18Rβ) 的共受体被募集形成高亲和力复合物[1]。IL-18/IL-18Rα/IL-18Rβ 三元复合物形成并置 IL-18Rα 和 IL-18Rβ 的细胞内 Toll-Interleukin-1 受体结构域。然后,接头分子骨髓分化因子 88 (MyD88) 可能在 TRAM 的帮助下被招募。MyD88 进一步与 IL-1 受体相关激酶 (IRAK) 4 和 IRAK1/2 相互作用,形成称为 Myddosome 的大分子组装体,随后通过 TRAF6 激活 IKK。最后,信号激活 NF-κB 和丝裂原激活蛋白激酶通路7,从而上调各种炎症细胞因子的表达。

生物活性
Measured by its ability to inhibit the IL-18- induced response of KG‑1 human acute myelogenous leukemia cells in the presence of Recombinant Human IL‑18 R alpha /IL‑1 R5. The ED50 for this effect is 2‑6 µg/mL.

This protein is an accessory subunit of the heterodimeric receptor for interleukin 18 (IL18), a proinflammatory cytokine involved in inducing cell-mediated immunity. This protein enhances the IL18-binding activity of the IL18 receptor and plays a role in signaling by IL18. Mutations in this gene are associated with Crohn's disease and inflammatory bowel disease, and susceptibility to celiac disease and leprosy. Alternatively spliced transcript variants of this gene have been described, but their full-length nature is not known.
